Universal Reaction Kit for Sequencing (Single Molecule Direct Sequencing) is designed for single molecule DNA sequencer. The kit includes a group of buffers and a piece of flow cell. Buffer groups are mainly polymerase buffer, plymerase, virtual terminator, imaging buffer, cleave reagent, cap reagent, rinse buffer and buffer H. Each component is for extending terminator, capturing image, cleaving bolcking site, protecting cleaved nucleotides, and washing chip surface, respectively. The flow cell is made of two pieces of glass. One piece of glass is modified with specific chemical which could capture DNA samples for sequencing and is divided to 16 channels. Two prepared DNA library could be hybridized to one channel. Each reaction buffer will be pumped in and out of channels through sequncer controller to finish sequencing reactions.
