It is used for staining of peroxidase-positive leukocytes in semen. Positive peroxidase in polymorphonuclear leukocytes can release oxygen from hydrogen peroxide to oxidize benzidine, which can then form a brown substance to deposit at the location of the peroxidase in the cytoplasm,thus to distinguish leukocytes from other round cells.
